The script states this question. It was not tested here: 1. Is PDE10A loss in HD striatum a driver of MSN dysfunction or a downstream consequence? 2. Do PDE10A inhibitors restore DARPP-32/CREB/PGC-1α signaling in HD preclinical models? 3. What is the presymptomatic therapeutic window for PDE10A targeting in HD? 4. How does the cAMP/cGMP axis interact with existing hypotheses (h020 MSN excitability, h021 NAD+/SIRT1, h037 BDNF/TrkB)? 5. Can PDE10A PET imaging serve as a progression biomarker for HD clinical trials?
